Description

UASp regulatory sequences drive expression of the dimerisation (DD), FH1 and FH2 domains of dia. The protein is tagged at the N-terminal end with EGFP (the tag is joined to the dia coding region via a HRYTSLYKKAGSAAAPFT linker).

Expression of diaDDFH1FH2.Scer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4en-e16E does not induce significant numbers of ectopic filopodia at the leading edge and there is little effect on protrusions of epidermal cells ventral to the leading edge. Lamellipodial area and filopodial number are significantly increased in leading edge cells. Filopodial lifetime is significantly increased compared to wild type.
