F532L
The nucleotide change was not reported and the position of the mutation is ambiguous. The TTT (Phe) codon may have changed to either CTT (Leu) or TTA/G (Leu). The annotated position is the first base of the codon.
aPKCts/aPKCk06403 flies are viable at the permissive temperature (25[o]C) and adult flies appear morphologically normal. Germline cyst encapsulation phenotypes are seen immediately after the mesenchymal-to-epithelial transition in region three of the germarium. The resulting follicular epithelium adherens junctions, apicobasal polarity and actin-myosin cytoskeleton appear normal. At the restrictive temperature (30[o]C) very few flies eclose from the pupal cases, with occasional escaper flies that exhibit abdominal dorsal closure defects. At the semi-permissive temperature (27-28[o]C) viability is variable, but almost all flies show significant dorsal closure defects.
aPKCts/Df(2R)l4 flies are viable at the permissive temperature (25[o]C) and adult flies are morphologically normal. However, females are sterile, with no larvae hatching from laid eggs even when crossed with wild type males. At the restrictive temperature (30[o]C) very few flies eclose from the pupal cases, with occasional escaper flies. At the semi-permissive temperature (27-28[o]C) viability is variable, but almost all flies show significant abdominal dorsal closure defects. Cyst encapsulation phenotypes are seen in aPKCts/Df(2R)l4 mutant ovaries at the permissive temperature.
aPKCts/aPKCk06403 third instar larval wing discs show a temperature sensitive induction of apoptosis. Whereas at the permissive temperatures (18[o]C and 25[o]C) there are low levels of cell extrusion and apoptosis, at the restrictive temperature (30[0]C) significantly higher levels of cell extrusion and apoptosis are seen, and the wings discs appear smaller than in controls. No phenotypes are seen in the heterozygous controls.

Much of the apoptosis is found in the basal region of the disc. No phenotypes are seen in the heterozygous controls.
At both the permissive and restrictive temperatures there is a significant decrease in the proportion of aPKCts/aPKCk06403 mutant wing disc cells with the correct planar spindle orientation compared to controls. The decrease is greater at 30[0]C than at 25[o]C.

Large aPKCts wing disc clones induced in first instar larvae show temperature-sensitive apoptosis.
Embryos maternally mutant for aPKCts fail to complete germ band extension (GBE), with a complete loss of epithelial integrity at the permissive temperature. Embryos develop normally until the onset of gastrulation and germ band extension.
Embryos maternally mutant for aPKCts/aPKCk06403 fail to complete germ band extension (GBE), with a complete loss of epithelial integrity at the permissive temperature.
Somatic follicular cell clones mutant for aPKCts show temperature-sensitive cyst encapsulation defects.

aPKCts/Df(2R)l4 has centrosome | maternal effect | embryonic stage phenotype, suppressible by par-1HMS00405/Scer\GAL4VP16.mat.αTub67C
aPKCts/aPKCk06403 has wing disc | third instar larval stage | heat sensitive phenotype, suppressible by raps[+]/pins193
aPKCts/aPKCk06403 has embryo | dorsal closure stage | heat sensitive phenotype, suppressible by raps[+]/pins193
aPKCts/Df(2R)l4, Scer\GAL4VP16.mat.αTub67C, par-1HMS00405 has centrosome | increased number | maternal effect | embryonic stage phenotype

Maternal expression of par-1HMS00405 under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 rescues the abnormal chromosome positioning of embryos from aPKCts/Df(2R)l4 mothers, but also results in enlarged cells with multiple centrosomes.
raps193 dominantly suppresses the temperature sensitive induction of apoptosis seen in aPKCts/aPKCk06403 third instar larval wing discs at the semi-permissive temperature (28[o]C). However, this suppression does not occur at the restrictive temperature (30[o]C).

Expression of BacA\p35Scer\UAS.cHa under the control of Scer\GAL4sd-SG29.1 at the permissive temperature (25[o]C) induces the development of large tumour-like tissues in the basal region of aPKCts/Df(2R)l4 mutant larval wing discs. Expression of BacA\p35Scer\UAS.cHa does not induce tumors in heterozygous controls.
