Description

Consists of two copies of the w exon 3 in opposite orientation to each other and separated by a w intron. The construct is terminated with a Hsp70 terminator. The resulting transcript after intron splicing, is predicted to create a perfect hairpin dsRNA of length 629 bp.

The GMR promoter drives expression of a tail-to-tail w repeat. The repeat comprises the 629bp third exon of w ligated to the 74bp second intron of w, ligated to another copy of the w third exon in the opposite orientation to the first copy.

Expression of wdsRNA.GMR/+ leads to pale orange eye color while expression of wdsRNA.GMR/wdsRNA.GMR leads to almost white eye color.

Two copies of the P{GMR-w.IR} transgene leads to an almost complete loss of eye pigmentation, while a single copy results in a less stringent eye phenotype, with a light yellow or orange eye color.

wdsRNA.GMR produces white eyes in a wild-type genetic background.

Flies carrying wdsRNA.GMR show a 90-fold reduction in drosopterin pigment levels compared to wild type.

Flies expressing two copies of wIR.GMR exhibit white eyes due to complete silencing of the w gene.

Expression of wdsRNA.GMR suppresses eye pigmentation.

The silencing of the w gene seen in flies carrying P{GMR-wIR} is less efficient if the flies are also homozygous for loqsf00791; flies carrying two copies of P{GMR-wIR} and homozygous for loqsf00791 do not have completely white eyes (in contrast to flies carrying two copies of P{GMR-wIR} in a wild-type background).

Dcr-1Q1147X, wIR.GMR animals have eyes that have reduced pigment, and are also reduced in size. The organisation of ommatidial facets is also disrupted and sensory bristles are missing over the eye surface. Other bristles, which flank the eye surface, are sometimes absent or exhibit hyperplasia.
