Expression of piwiHMS00606 under the control of Scer\GAL4GMR.PU does not result in any obvious phenotype in the adult eye or third instar larval wing disc, as compared to controls.

The expression of piwiHMS00606 under the control of Scer\GAL4C587 at room temperature leads to germarium defects as compared to controls: germ line stem cells range from completely absent to increased in number, some locating away from cap cells; escort cells are severely decreased in number, show increases in DNA damage and in apoptosis, and lose their long cellular processes. Similar germ line cell defects are observed in adults kept at 18[o]C throughout development and shifted to 29[o]C upon eclosion, but not in adults kept continuously at 18[o]C throughout development and adulthood.

The expression of piwiHMS00606 under the control of Scer\GAL4VP16.nos.UTR results in moderate to severe decreases in the number of primordial germ cells in the third instar larval female gonad and results in a complete loss of germ line stem cells in the germarium of newly eclosed adults, as compared to controls.

The clonal expression of piwiHMS00606 under the control of Scer\GAL4VP16.nos.-FRT in the germarium leads to the progressive loss of germline stem cells within the clones (a significant proportion of germaria lack germ line stem cells 3 weeks after clone induction, but not 1 week after clone induction), as compared to controls; conversely, some of these germaria accumulate excessive germline stem cells outside of the clone region.

Females expressing piwiHMS00606 under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 do not show gross morphological defects in the ovary, but are sterile.
