Expression of tshJF02856 in the fat body under the control of Scer\GAL4DcG results in significant hyperglycaemia.

Only 12% of animals expressing tshJF02856 under the control of Scer\GAL4tsh-c724 emerge as adults, and the adults ares short lived compared to controls. 1 week old adults have grossly distended abdomens due to fluid retention.

Malpighian tubules from adults expressing tshJF02856 under the control of Scer\GAL4tsh-c724 show significantly lower basal secretion rates than control tubules. Addition of cAMP increases the secretion rate in the mutant tubules, however the peak rate is significantly lower than control tubules. Addition of Leucokinin in addition to cAMP has no further stimulatory effect on the mutant tubules, in contrast to controls which show a further doubling of the secretion rate.

Uric acid crystals accumulate in the lumen of the Malpighian tubules in animals expressing tshJF02856 under the control of Scer\GAL4tsh-c724.

The number of stellate cells in the anterior and posterior Malpighian tubules is normal in adults expressing tshJF02856 under the control of Scer\GAL4tsh-c724 and the spacing of stellate cells in these tubules is indistinguishable from wild type. The morphogenetic transformations that usually occur in the stellate and bar-shaped cells of the Malpighian tubules during pupation do not occur in the mutant animals; the cells remain cuboidal both in the main and initial/transitional regions of the adult tubules. The mutant adult stellate cells have less than half the surface area of stellate cells in wild-type tubules.

Co-expression of tioScer\UAS.cLa restores stellate cell morphology is restored in adults expressing tshJF02856 under the control of Scer\GAL4tsh-c724. In addition, adult viability is partially rescued (22% of adults eclose) and the adults that emerge do not have bloated abdomens.
