UASt regulatory sequences drive expression of an inverted repeat.
Expression of PbursGD6814 under the control of Scer\GAL4nSyb.PU (in combination with UAS-Dicer2 to increase RNAi efficiency) results in a significant increase in the latency to sperm ejection in females after the end of copulation.
In contrast to wild type flies placed in a confined environment, newly emerged flies expressing PbursGD6814 under the control of Scer\GAL4Burs.PP do not show wing expansion upon stimulation of the CCAP-expressing neurons. Approximately 30% of the mutant flies do show abdominal flexion (a behavioural signature of wing expansion), but only after environmental delays similar to those of flies in which the CCAP-expressing neurons are not activated.
Newly emerged flies expressing PbursGD6814 in the B[[AG]] neurons (the intersection of the Hsim\VP1699 and Scer\GAL4DBD.Burs.T:Zzzz\Zip- split-gal4 hemidrivers) and placed in a confined environment still show the abdominal flexion behaviours characteristic of wing expansion, although the wings typically undergo little to no expansion during the abdominal flexion.
Expression of pbursGD6814 under the control of Scer\GAL4dpp.blk1 results in multiple cellular hairs in the wing.
Expression of pbursGD6814 under the control of Scer\GAL4en-e16E results in multiple cellular hairs in the wing.
Expression of pbursGD6814 under the control of Scer\GAL4ap-md544 results in multiple cellular hairs in the wing.
Adults expressing PbursGD6814 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
Dcr-2UAS.cDa, PbursGD6814, Scer\GAL4hs.2sev has abnormal planar polarity phenotype
Dcr-2UAS.cDa, PbursGD6814, Scer\GAL4hs.2sev has ommatidium phenotype
Co-expression of pbursGD6814 with Dcr-2Scer\UAS.cDa under the control of Scer\GAL4hs.2sev results in ommatidial planar cell polarity (PCP)-defects in the eye.
