Description

Imprecise excision of the P{Mae-UAS.6.11} element, which results in deletion of 2530bp of the upd2 genomic region, from 2264bp upstream of the AUG codon to 266bp downstream. This removes the 5'UTR and coding sequence for the first 89 amino acid residues of the protein product.

Comment:

A 2530 bp deletion resulting from the imprecise excision of P{Mae-UAS.6.11}upd2GG01031, which extends from 2264bp upstream of the upd2 AUG codon to 266bp downstream.

Detailed Description

Hemizygous adult males appear smaller and slimmer than wild-type controls. Wing area is reduced by 10% overall and there is a significant reduction in both cell number and cell size compared to wild type.

Mutant adults show a significant reduction in triacylglycerol levels and a significant increase in circulating glucose levels compared to controls.

Mutant adults do not show defects in feeding behaviour.

Mutant larvae show a striking accumulation of lipid droplets in oenocytes under fed conditions, in contrast to wild-type controls.

Expression of upd1dsRNA.Scer\UAS.cUa partially suppresses the overgrowth and invasion of the nerve cord seen in l(2)glunspecified mutant eye-antennal disc clones that simultaneously express Ras85DV12.Scer\UAS under the control of Scer\GAL4Act.PU. The phenotype is further suppressed when the clones are generated in a upd2Δ3-62 mutant background.

upd2Δ3-62 does not suppress the overgrowth and invasion of the nerve cord seen in l(2)glunspecified mutant eye-antennal disc clones that simultaneously express Ras85DV12.Scer\UAS under the control of Scer\GAL4Act.PU.

Expression of Bhal\NaChBacScer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4Ilp3.PB rescues the reduced wing area of upd2Δ3-62 mutants.

Expression of ShEKO.Scer\UAS.T:Avic\GFP-GL under the control of Scer\GAL4Ilp3.PB does exacerbate the reduced wing area phenotype of upd2Δ3-62 mutants.

Expression of Hsap\LEPScer\UAS.cRa under the control of Scer\GAL4Cg.PA rescues the reduced wing area and reduced triacylglycerol levels seen in upd2Δ3-62 flies.

Complementation and Rescue Data
Comments

The reduction in wing size and reduction in triacylglycerol levels seen in upd2Δ3-62 flies is rescued by expression of upd2Scer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4Cg.PA.
