Mi{ET1}VGATMB01219 is inserted approximately between nucleotides 286 and 287 of the VGAT coding sequence, within the second exon.
VGATMB01219 flies, expressing VGATScer\UAS.cFa under the control of Scer\GAL4VGAT and Scer\GAL80ts.αTub84B, to allow rescue past the embryonic lethality to the adult stage do not show any gross anatomical abnormalities in the head or nervous system.
VGATMB01219 mutant flies, expressing VGATScer\UAS.cFa under the control of Scer\GAL4VGAT and Scer\GAL80ts.αTub84B, to allow rescue past the embryonic lethality to the adult stage, exhibit normal ranges in wing beat frequency and amplitude. These flies do not exhibit differences in optomotor control capacity compared to controls.
VGATMB01219 mutant flies, expressing VGATScer\UAS.cFa under the control of Scer\GAL4VGAT and Scer\GAL80ts.αTub84B, show a reduced ability to fixate a stripe moving against a uniform white background and a diminished capacity for tracking a stripe moving against a counter-rotating wide-field background in which any displacement of the stripe is matched exactly by motion of the background in the opposite direction. General wide-filed optomotor function is intact in these mutants, while the behavioral deficit in these flies is specific for figure detection computations required to follow or fixate the direction of gaze on a small-field object, and more so for the control of response magnitude rather than timing.
VGATMB01219 is rescued by Scer\GAL4VGAT.PF/VGATUAS.cFa
