Expression of MbsScer\UAS.cLa under the control of Scer\GAL4P0.5.Pdf results in an increase in axonal volume and spread of s-LNv neuron projections compared to controls when expression is induced at ZT24 (dawn) and spread is measured 12 hours later.

In contrast to controls, which adjust their morning and evening activity peaks to align with seasonal dawn and dusk, flies expressing MbsScer\UAS.cLa under the control of Scer\GAL4P0.5.Pdf (restricted to the adult stages using Scer\GAL80ts.αTub84B) are active later than control flies when exposed to a summer light:dark cycle (14 hours day:10 hours night). Timing of activity is unaffected compared to controls when the flies are exposed to a winter cycle (10 hours day:14 hours night). Normal locomotor activity is seen when the flies are kept in complete darkness (DD).

Co-expression of MbsScer\UAS.cLa suppresses the reduction in axonal volume and spread of s-LNv neuron projections seen at ZT24 (dawn), following 12 hours of expression of Rho1Scer\UAS.cMa under the control of Scer\GAL4P0.5.Pdf (restricted to the adult stages using Scer\GAL80ts.αTub84B).

The behavioural arrhythmicity of flies expressing Rho1Scer\UAS.cMa under the control of Scer\GAL4P0.5.Pdf from birth is rescued upon co-expression of MbsScer\UAS.cLa.

Expression of MbsScer\UAS.cLa, driven by Scer\GAL4da.G32, partially rescues the embryonic lethality of MbsT541/MbsT791 transheterozygotes, allowing over 90% of these mutants to survive to the pupal stage and 45% to survive to adulthood. Likewise, expression of the transgene allows 80% of MbsT541/MbsT666 mutants to survive to the pupal stage, but only 6% to survive to adulthood. MbsT666 mutant eye disc clones that express MbsScer\UAS.cLa under the control of Scer\GAL4tub (generated using the MARCM technique) show normal apical localization within the eye disc, thus rescuing the eye development phenotype of the MbsT666 clones.
