Scer\GAL4bi-omb-Gal4-driven expression of midScer\UAS.cBa results in ectopic sex combs and transverse bristle rows in the dorsal basitarsus and distal tibia of all male first legs.
In the second leg, ectopic expression of Scer\GAL4bi-omb-Gal4-driven midScer\UAS.cBa in the dorsal tibia results in ectopic bristles similar to the apical and spur bristles. Scer\GAL4Act5C.PI-driven expression of midScer\UAS.cBa in small somatic clones also results in the appearance of similar ectopic bristles. Small clones of midScer\UAS.cBa-expressing cells either in or adjacent to rows 2/7 and 3/6 induce ventral row 1/8 bristles cell autonomously.
Ventrolateral or dorsolateral Scer\GAL4Act5C.PI-driven midScer\UAS.cBa-expressing clones in the tarsus are often transformed to ventral character.
By stage 17, phenotypic effects of ectopic midScer\UAS.cBa expression in the mesoderm, driven by Scer\GAL4twi.PB, can be observed; dorsal vessel development is disrupted and in some embryos the two halves of the heart fail to fuse. The same phenotype is observed when midScer\UAS.cBa is expressed in the dorsal mesoderm and ectoderm under the control of Scer\GAL4pnr-MD237. Expression of midScer\UAS.cBa throughout the mesoderm, under the control of Scer\GAL4how-24B, causes the ectopic formation of up to 30 extra cardioblasts per half dorsal vessel, but only rarely disrupts the fusing of the two halves of the heart.
Expression of midScer\UAS.cBa under the control of either Scer\GAL4ptc-559.1 or Scer\GAL4sca-P309 results in nearly all naked cuticle being replaced by denticles in the posterior half of the larva. Larvae expressing midScer\UAS.cBa under the control of Scer\GAL4ptc-559.1 have a reduced body size, strong segmental indentations and head malformations. Expression of midScer\UAS.cBa under the control of Scer\GAL4prd.RG1 results in larvae with fusion of alternating denticle belts.
Scer\GAL4prd.RG1, midUAS.cBa has ventral denticle belt phenotype, suppressible by wgwg.3'UTR.UAS/Scer\GAL4prd.RG1
Scer\GAL4twi.PG, lolaC28/lola22.05, midUAS.cBa has fat body/gonad primordium phenotype
Scer\GAL4twi.PG, lolaC28/lola22.05, midUAS.cBa has gonad | embryonic stage phenotype
Scer\GAL4twi.PG, lolaC28/lola22.05, midUAS.cBa has gonadal sheath proper primordium | increased number phenotype
Mmus\Lef1UAS.Tag:T7, Scer\GAL4Act5C.PI, midUAS.cBa has metathoracic tarsal bristle phenotype
Mmus\Lef1UAS.Tag:T7, Scer\GAL4Act5C.PI, midUAS.cBa has prothoracic tarsal bristle phenotype
When midScer\UAS.cBa is expressed in the mesoderm of lolaC28/lola22.05 mutant embryos under the control of Scer\GAL4twi.PG 'super-elongated' gonads are produced in more than 50% of stage 15 embryos. Although somatic gonadal precursor (SGP) cluster formation occurs, these gonads remain elongated, spanning several parasegments. Far more SGPs are seen than in sibling controls, and these extra SGPs arise from conversion from fat body cells at stage 13.
Co-expression of wgwg.3'UTR.Scer\UAS results in near complete suppression of the fusion of alternating denticle belts seen in larvae expressing midScer\UAS.cBa under the control of Scer\GAL4prd.RG1.
Clones expressing both midScer\UAS.cBa and Mmus\Lef1Scer\UAS.cRa in the tarsus driven by Scer\GAL4bi-omb-Gal4 are recovered more often in lateral and dorsal cells, indicating altered sorting behaviour. Dorsolateral clones expressing both midScer\UAS.cBa and Mmus\Lef1Scer\UAS.cRa do not transform towards ventral fate, whereas ventrolateral clones do so sometimes.
Scer\GAL4twi.PG/midUAS.cBa partially rescues midB23/mid1
Scer\GAL4Six4.PT/midUAS.cBa partially rescues midB23/mid1
Expression of midScer\UAS.cBa in the mesoderm under the control of Scer\GAL4twi.PG fully rescues the lack of somatic gonadal precursor (SGP) cluster fusion and germ cell ensheathment seen in midB23/mid1 mutant embryos. The number of scattered germ cells is reduced. The SGP-related defects are also rescued in the majority of embryos when midScer\UAS.cBa is expressed under the control of Scer\GAL4Six4.PT, but not to the extent seen with Scer\GAL4twi.PG.
