    Description

BruceE81 contains an in frame deletion that removes the BIR domain.

Deletion of 534 bp in-frame = codons 234-411 (predicted to encode the BIR domain).

    Detailed Description
    Statement
    Reference

Mutant females have a significant increase in the number of degenerating egg chambers during mid-oogenesis compared to wild type.

Well-fed BruceE81 mutant flies display increased levels of autophagy (punctate Lysotracker staining) in region two of the germarium and in stage eight degenerating egg chambers compared to controls.

In contrast to wild type, the ovaries of well-fed BruceE81 mutant flies contain a large number of degenerating stage eight egg chambers.

An increased number of apoptotic (TUNEL positive) cells are seen in region two cysts in well-fed BruceE81 mutant flies than are seen in wild type. TUNEL positive cells are also seen in BruceE81 mutant stage eight egg chambers.

BruceE81 homozygotes are viable but male-sterile. Mutant spermatid nuclei are significantly more condensed and rounded than wild-type, and are scattered throughout the cyst. In addition, there are fewer nuclei, and many nuclei are stained faintly, presumably because they undergo degeneration. Although an indivdualisation complex forms, it is highly reduced and scattered.

    Genetic Interactions
    Statement
    Reference

A BruceE81 heterozygous background dominantly enhances the rprGMR.PD eye ablation phenotype.

A BruceE81 heterozygous background does not affect the pigment cell death phenotype caused by the overexpression of Ncpro.Scer\UAS in the developing eye under the control of Scer\GAL4GMR.PF.

A BruceE81/+ heterozygous background has no effect on the eye ablation phenotype found upon co-expression of ArkScer\UAS.T:Zzzz\His6,T:Hsap\MYC and Ncpro.Scer\UAS in the developing eye under the control of Scer\GAL4GMR.PF.
