FlyBase curator comment: Interactions with Dg[086] and/or Dg[323] were detected in a Dg[086]/+ or Dg[323]/+ background which exhibits no obvious changes in muscle morphology. Nonetheless, these interactions have been captured as 'modifier' ('exacerbates') annotations here to best capture the experimental finding and the authors' intention.

dyscc03838/dyscc03838 and dyscs168/dyscc03838 third instar larvae show a significant reduction in bouton number (both type 1b and type 1s boutons) and a significant increase in the size of type 1s boutons at the neuromuscular junction.

Homozygous larvae show a significant increase in the rate of body wall peristalsis compared to controls.

Mutant larvae show a significant increase in evoked excitatory junctional current (EJC) amplitude and width compared to controls. There is a significant increase in spontaneous miniature EJC frequency compared to controls.

Most dyscc03838 homozygotes and dyscc03838/Df(3L)BSC614 animals display arrhythmic locomotor patterns in constant dark conditions.

In contrast to wild type flies, dyscc03838 homozygotes do not exhibit anticipation of lights-on in 12 h light:dark conditions, while anticipation of lights-off is maintained. Overall daytime and nighttime activity is greater in these mutants compared to wild type in these conditions.

dyscs168/dyscc03838 animals display arrhythmic locomotor patterns in constant dark conditions.

dyscc03838/dyscc05107 animals display arrhythmic locomotor patterns in constant dark conditions.

Bouton size at the neuromuscular junction in dyscc03838 sloMB11481 double mutant larvae is significantly increased compared to wild type, but is not significantly different from that seen in dyscc03838 single mutants. The reduction in bouton number at the neuromuscular junction seen in dyscc03838 larvae is suppressed in dyscc03838 sloMB11481 double mutant larvae.

Evoked excitatory junctional currents in dyscc03838 sloMB11481 double mutant larvae have the same phenotypes as in either single mutant. Miniature EJC frequency in dyscc03838 sloMB11481 double mutant larvae is not significantly different from that in either single mutant.

dysc Dys double heterozygous flies (dyscc03838/Df(3R)Exel6184) exhibit indirect flight muscle degeneration.

dyscc03838 DgO86 double heterozygous flies exhibit indirect flight muscle degeneration.

One copy of dyscc03838 enhances the indirect flight muscle degeneration seen when DysdsRNA.NH2.Scer\UAS is expressed under the control of Scer\GAL4Act.PU.

One copy of dyscc03838 enhances the indirect flight muscle degeneration seen when DgdsRNA.Scer\UAS is expressed under the control of Scer\GAL4tub.PU.

dyscc03838 Dg323 double heterozygous flies exhibit indirect flight muscle degeneration.

Comments

The increase in evoked excitatory junctional current amplitude seen in dyscc03838 larvae is rescued by expression of dyscScer\UAS.long under the control of Scer\GAL4elav.PU.

Scer\GAL4elav.PU-, Scer\GAL4dysc.PJ-, or Scer\GAL4C164-mediated expression of dyscScer\UAS.g rescues the locomotor rhythm defects of dyscc03838/dyscs168 animals.

Scer\GAL4elav.PU-mediated expression of dyscScer\UAS.f does not rescue the locomotor rhythm defects of dyscc03838/dyscs168 animals.
