Expression of SdcScer\UAS.cJa driven post-synaptically by Scer\GAL4how-24B or presynaptically by Scer\GAL4elav.PU leads to a significant increase in the number of boutons per neuromuscular junction, compared to controls.

Expression of SdcScer\UAS.cJa under the control of either Scer\GAL4how-24B or Scer\GAL4elav.PU results in an increase in bouton number at the neuromuscular junction compared to wild type.

Expression of SdcScer\UAS.cJa, under the control of Scer\GAL4how-24B, leads to SNa bifurcation with a penetrance of 18%. No ISNb phenotype is observed.

Sdc10608/Df(2R)48 third instar larvae (in which Sara function has been rescued by SaraUbi.PJ) have a decrease in the number of boutons per neuromuscular junction, compared to controls; this phenotype is completely rescued by pre- (Scer\GAL4elav.PU) or post- (Scer\GAL4how-24B) synaptic expression of SdcScer\UAS.FL.T:Hsap\MYC, or pre- (and partial rescue with post-) synaptic expression of SdcScer\UAS.cJa.

Larvae co-expressing SdcScer\UAS.cJa and dlpScer\UAS.cBa under the control of Scer\GAL4how-24B show a reduction in bouton number at the neuromuscular junction compared to wild type, as occurs in larvae expressing dlpScer\UAS.cBa alone under the control of Scer\GAL4how-24B.

When SdcScer\UAS.cJa is expressed, under the control of Scer\GAL4how-24B, in a Lar13.2/Lar5.5 background, the SNa bifurcation phenotype seen when the transgene is expressed in a wild-type background is suppressed.

Comments

Sdc10608/Df(2R)48 third instar larvae (in which Sara function has been rescued by SaraUbi.PJ) have a decrease in the number of boutons per neuromuscular junction, compared to controls; this phenotype is completely rescued by pre- (Scer\GAL4elav.PU) and partial rescue with post- (Scer\GAL4how-24B) synaptic expression of SdcScer\UAS.cJa.

Expression of SdcScer\UAS.cJa under the control of Scer\GAL4elav.PU almost completely rescues the reduction in bouton number at the neuromuscular junction which is seen in Sdc10608/Df(2R)48 larvae (carrying SaraUbi.PJ to provide Sara function).

Expression of SdcScer\UAS.cJa under the control of Scer\GAL4how-24B provides limited rescue of the reduction in bouton number at the neuromuscular junction which is seen in homozygous Sdc10608 larvae.

Expression of SdcScer\UAS.cJa under the control of Scer\GAL4sli.PS fails to rescue the axon midline crossing phenotype seen in Df(2R)48 embryos in which Sara function has been rescued by SaraUbi.PJ. Expression of SdcScer\UAS.cJa under the control of Scer\GAL4elav.PLu rescues the axon midline crossing phenotype seen in Df(2R)48 embryos in which Sara function has been rescued by SaraUbi.PJ.
