Expression of nwkScer\UAS.cCa under the control of Scer\GAL4elav-C155 does not affect synaptic growth at the larval neuromuscular junction (both the average total number of boutons and the number of satellite boutons are normal in the mutant larvae).

Expression of nwkScer\UAS.cCa under the control of Scer\GAL4elav-C155 has no effect on synaptic growth or bouton number in third instar larvae.

Co-expression of nwkScer\UAS.cCa with Atg1Scer\UAS.cSa, under the control of Scer\GAL4elav.PLu fails to suppress Atg1Scer\UAS.cSa-mediated neuromuscular junction overgrowth.

Co-expression of nwkScer\UAS.cCa suppresses the increase in the total number of boutons and in the number of satellite boutons at the neuromuscular junction that is seen in larvae expressing two copies of tkvQ199D.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4elav-C155.

Expression of nwkScer\UAS.cCa under the control of Scer\GAL4elav-C155 suppresses the increase in the total number of boutons and in the number of satellite boutons at the neuromuscular junction that is seen in homozygous Dadj1E4 larvae.

Comments

Expression of nwkScer\UAS.cCa under the control of Scer\GAL4elav-C155 partially rescues the synaptic overgrowth and increased bouton number phenotypes seen in nwk1/nwk2 third instar larvae.

The temperature sensitive paralysis phenotype of nwk2 adults is rescued by expression of nwkScer\UAS.cCa under the control of Scer\GAL4elav-C155; after 15 minutes at 28oC, more than 90% of the rescued adults continue to walk and climb normally. Expression of nwkScer\UAS.cCa under the control of Scer\GAL4elav.PLu partially rescues the neuromuscular junction (NMJ) phenotypes seen in nwk2 larvae; the normalised numbers of boutons and branches are fully rescued at NMJ 6/7 and NMJ 4, and the increase in NMJ length seen in nwk2 larvae is substantially rescued. The increased incidence of hyperbranched boutons per NMJ seen in nwk2 larvae is not altered in the rescued larvae. Excitatory junctional current (EJC) amplitude is fully rescued and miniature EJC amplitude is partially rescued.
