Third instar larval eye-antennal disc clones expressing banC.Scer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4Act5C.PI exhibit reduced epithelial folding and abnormal mixing of cells across the field boundary at the A1 fold, as compared to controls.

Larval eye-antennal discs expressing banC.Scer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4dpp.blk1 exhibit decreased cell proliferation, but no significant change in apoptosis levels, as compared to controls.

Expression of banC.Scer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4esg-NP5130 increases the number of midgut precursor cells. An increased number of intestinal stem cells are undergoing DNA replication compared to controls.

Expression of banC.Scer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4esg-NP5130 (restricted to the adult stages using Scer\GAL80ts.αTub84B) results in an increase in midgut precursor cells compared to controls. An increase in cell proliferation (pH3 staining) is observed.

Intestinal stem cell differentiation is unaffected in clones expressing banC.Scer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL80ts.αTub84B, but the clones exhibit a growth advantage over control clones.

Expressing mir-banC.UAS.EGFP under the control of Scer\GAL4ap-md544 (in combination with a Gal80[ts] transgene to inhibit expression until early third instar) results in severely overgrown wing discs in the dorsal compartment, but with an epithelial organization similar to wild-type controls in third instar larvae. There is no effect on cell death.

Late third instar wing discs expressing banC.Scer\UAS.T:Avic\GFP-EGFP under the control of Scer\GAL4sd-SG29.1 have greatly disturbed dorsal-ventral (DV) boundaries compared to controls. The anterior-posterior boundary remains straight and smooth. BrdU staining shows that increased boundary cell proliferation is occurring at the DV boundary. This uneven DV boundary phenotype is also observed in the wing discs of mid-third instar larvae, and when banC.Scer\UAS.T:Avic\GFP-EGFP is expressed under the control of Scer\GAL4vg.int2.1.

When banC.Scer\UAS.T:Avic\GFP-EGFP is driven by Scer\GAL4en-e16E overgrowth is seen in the posterior compartment of the wing and of segments in the larval body. banC.Scer\UAS.T:Avic\GFP-EGFP when driven by Scer\GAL4ptc-559.1 is sufficient to direct cells in the non-proliferating zone of the wing disc to enter S phase.

Clones both homozygous for Rab5LL00467 and expressing mir-banC.UAS.EGFP under the control of Scer\GAL4Act5C.PI form tumors in the larval brain (which metastasize to the ventral nerve cord). Larval eye/antennal disc clones also form tumors and their induction leads to lethality. Co-expression of JhI-21HMS02271 suppresses tumorigenesis and lethality, but adults exhibit an eye phenotype.

Co-expressing EgfrUAS.cBa and mir-banC.UAS.EGFP under the control of Scer\GAL4ap-md544 (in combination with a Gal80[ts] transgene to inhibit expression until early third instar) results in severely overgrown and deformed wing discs with a disrupted epithelial organization (resembling tissues undergoing a neoplastic transformation) in third instar larvae.

Expression of dmGD1419 is unable to suppress the dorsal-ventral boundary defects seen when banC.Scer\UAS.T:Avic\GFP-EGFP is expressed under the control of Scer\GAL4sd-SG29.1.

Expression of trblScer\UAS.cMa is unable to suppress the dorsal-ventral boundary defects seen when banC.Scer\UAS.T:Avic\GFP-EGFP is expressed under the control of Scer\GAL4sd-SG29.1.

The addition of banC.Scer\UAS.T:Avic\GFP-EGFP (driven by Scer\GAL4arm.PS) to Df(3L)banΔ1 animals, rescues the growth defect in the imaginal discs and leads to morphologically normal pupae, many of which survive to adulthood.
