Description

P{EP} insertion 643bp upstream of the Hsp22 translation initiation codon. The P{EP} element is oriented such that expression from the element should result in overexpression of Hsp22.

The P{EP}Hsp67BbEP3247 insertion is within a dicistronic transcript which encodes two open reading frames, represented by Hsp22 and Hsp67Bb. The insert is oriented in the same direction as the direction of transcription and is downstream of the Hsp67Bb open reading frame and upstream of the Hsp22 open reading frame.

Animals expressing Hsp22EP3247 under the control of Scer\GAL4Act5C.PI show a 29.2% increase in mean lifespan compared to controls, and the maximum lifespan of the mutant flies is approximately 115 days compared to approximately 95 days in controls.

Expression of Hsp22EP3247 under the control of Scer\GAL4Act is able to extend the mean life span under hyperoxic conditions by 15%.

Expression of Hsp22EP3247 under the control of Scer\GAL4ple.PF rescues the degeneration of dopaminergic neurons that is seen in flies under hyperoxia.
