Ras85Dx7b mutant eye disc clones in a minute background do not show any differentiation of ommatidial cells.

Ras85Dx7b homozygous clones in the dorsal air sac primordium grow more slowly than wild-type clones and never contribute to the tip of the primordium.

Ras85Dx7b clones induced in the wing disc in the first larval instar can be recovered in the prospective wing blade domain, wing hinge and medial notum, but are generally not seen in the prospective lateral notum domain.

Homozygous clones are not recovered in the adult eye. Homozygous clones are seen in the eye imaginal disc, but the size of the clone is reduced compared to the wild-type sister clone (twin spot). Clones in the eye behind the morphogenetic furrow are often wedge shaped and much reduced in size. Clusters of apoptotic cells are seen in clones posterior to the morphogenetic furrow. Homozygous clones in the eye which are Minute+ (in a RpS32 background) are substantially larger and cover large areas of the disc. As seen in a wild-type background, the size of these clones is reduced behind the morphogenetic furrow. Flies homozygous for Ras85Dx7b in the eye disc develop with a severely reduced head that almost completely lacks eyes. Introduction of Ras85DD38E into flies that are homozygous for Ras85Dx7b in the eye disc results in a slight enlargement of the eye field, due to the presence of unpigmented cuticle, but no additional ommatidial structures are formed.

rlSem/+ alone is not sufficient to rescue the reduced eye and head phenotypes of flies that are homozygous for Ras85Dx7b in the eye disc. However, the addition of Ras85DD38E in combination with rlSem results in a substantial rescue of the Ras85Dx7b eye and head size phenotypes; differentiated photoreceptor cells are seen and a substantial fraction of the ommatidia contain the full complement of photoreceptors.

Ras85DD38E partially rescues the growth defects of Ras85Dx7b clones in the eye disc. In addition, the rescued cells survive beyond the morphogenetic furrow and R8 photoreceptor cells are seen to develop in the clone. The partial rescue is not increased by increased Ras85DD38E copy number nor by the addition of Ras85DY40C.
