Ommatidia often under-rotate in animals expressing edScer\UAS.cBa under the control of Scer\GAL4hs.2sev.

Animals expressing edScer\UAS.cBa under the control of Scer\GAL4ro.PF show mostly normal rotation of the ommatidia. Some ommatidia are missing photoreceptors.

Expression of edScer\UAS.cBa under the control of Scer\GAL4GMR.PF results in severe disruption of adult eye tissue.

Expression of edScer\UAS.cBa under the control of Scer\GAL4twi.PG results in mild defects in lateral transverse muscle morphology.

The number of macrochaetae/heminotum in animals expressing edScer\UAS.cBa under the control of Scer\GAL4sca-C253 is - anterior+posterior notopleural: 1.23 (some flies lack macrochaetae at this position, others have duplications at this position), presutural: 0.90, anterior supraalar: 0.95, posterior supraalar: 0.88, anterior postalar: 0.98, posterior postalar: 1.00, anterior+posterior dorsocentral: 1.58 and anterior+posterior scutellar: 0.58. This represents a mild suppression of notum macrochaetae and the appearance of some extra macrochaetae in the notopleural position. The number of macrochaetae/heminotum in animals expressing edScer\UAS.cBa under the control of Scer\GAL4C-765 is - anterior+posterior notopleural: 2.25, presutural: 1.25, anterior supraalar: 1.15, posterior supraalar: 1.03, anterior postalar: 1.20, posterior postalar: 1.03, anterior+posterior dorsocentral: 2.85 and anterior+posterior scutellar: 3.70. Expression of edScer\UAS.cBa under the control of Scer\GAL4sca-537.4 results in the partial suppression of some macrochaetae (such as dorsocentral and scutellar) and some microchaetae, and the occasional duplication of some macrochaetae on the notum. Expression of edScer\UAS.cBa under the control of Scer\GAL4ap-md544 at 20oC results in the loss of macrochaetae at some positions and extra bristles at others on the notum. Expression of edScer\UAS.cBa under the control of Scer\GAL4Bx-MS1096 can result in extra macrochaetae on the notum.

When edScer\UAS.cBa is driven by Scer\GAL4GMR.PF, animals exhibit a rough eye phenotype and loss of photoreceptor and cone cells. On average 10-15% of ommatidia miss photoreceptor or cone cells.

edScer\UAS.cBa; Scer\GAL4sca-537.4 flies have many bristles missing. Examination of larval wing discs at puparium formation reveals that very few sensillum precursor cells have been selected.

When expression of one copy of edScer\UAS.cBa is driven by Scer\GAL4GMR.PF, eyes are small and rough with reduced number of photoreceptors. When expression of two copies of edScer\UAS.cBa is driven by Scer\GAL4GMR.PF, eyes fail to develop at all. When expression is driven by Scer\GAL4Bx-MS1096 wings are reduced in size and lack wing veins. When expression is driven by Scer\GAL4T155 or Scer\GAL4CY2 resulting eggs have completely normal dorsal appendages.

Expression of edScer\UAS.cBa, under the control of Scer\GAL4twi.PG, in a Gripex36 background, greatly enhances the muscle defects of Gripex36 embryos; ventrolateral muscles are more severely deranged and often appear to adhere to other muscles, whereas lateral transverse muscles split.

The addition of Nrg180.I.Scer\UAS enhances the rough eye phenotype seen in edScer\UAS.cBa, Scer\GAL4GMR.PF animals with a reduction in the number of ommatidia and a decrease in the number of bristles. In addition, a significantly higher percentage of ommatidia contain fewer photoreceptor and cone cells. The addition of Nrgl4/+ significantly suppresses the loss of cone cell phenotype seen in edScer\UAS.cBa, Scer\GAL4GMR.PF animals but has no effect on photoreceptor loss. The addition of Fas1::NrgGPI.Scer\UAS enhances the rough eye phenotype seen in edScer\UAS.cBa, Scer\GAL4GMR.PF.

Comments

Expression of edScer\UAS.cBa rescues the posterior wing overgrowth phenotype seen when edGD79 is expressed under the control of Scer\GAL4hh-Gal4.

Ubiquitous expression of edScer\UAS.cBa under the control of Scer\GAL4C-765 rescues the cell sorting phenotypes seen in ed1X5 mutant wing disc clones. The boundary between the clone and the surrounding wild type cells is no longer smooth edged, instead appearing jagged as in wild type. Normal levels of shg are seen.

The poor viability of ed1X5 homozygous clones in the wing disc is rescued if the clones are induced in a background where edScer\UAS.cBa is expressed under the control of Scer\GAL4C-765.
