P{EP} insertion in the 5' untranslated region.

P{EP} insertion upstream of trbl. The transcriptional orientation of the P{EP} element is the same as the direction of transcription of the trbl locus.

The P{EP} insertion is 8bp upstream of the presumed transcription start site of trbl.

Expression of trblEP3519 under the control of Scer\GAL4slbo.2.6 partially blocks border cell migration.

Expression of trblEP3519 under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 (the animals are initially raised at 18oC and are then shifted to 29oC after 7 days) causes spermatogonia to continue mitotic proliferation at the expense of differentiation. This defect is seen in at least 50% of testes examined.

Expression of trblEP3519 under the control of Scer\GAL4sca-537.4 results in the loss of macrochaetae. Sensory organs with no socket and one shaft and thickened shafts are seen.

Cells in the ventral domain (mitotic domain 10) are the first to enter mitosis in trblEP3519 embryos. The order of the other mitotic domains is not altered. As a consequence of the early mitosis in mitotic domain 10, the mesodermal precursors remain on the surface and do not form a proper ventral furrow.

Expression of trblEP3519 under the control of Scer\GAL4slbo.2.6 affects border cell migration.

At the time when the ventral furrow has invaginated fully in wild-type, in homozygous trblEP3519 mutant embryos, the majority of the prospective mesodermal cells are in mitosis (dividing too early) and no furrow is formed. In contrast to wild-type homozygous mutants approximately 75% of embryos have dividing pole cells between stage 6 and stage 10. Only individual cells are seen in mitosis. No phenotype is seen when trblEP3519 is driven by Scer\GAL4twi.PG, Scer\GAL4prd.RG1, Scer\GAL4Kr.PM, Scer\GAL4sd-SG29.1, Scer\GAL4ap-md544, Scer\GAL4hs.2sev, Scer\GAL4GMR.15 or Scer\GAL4GMR.12.
