Clones of ciScer\UAS.cDa in which expression is driven by Scer\GAL4hh-Gal4, generated along the A/P boundary of the wing disc using the MARCM technique, are misplaced into the A territory.

Cells in clones in the morphogenetic furrow in the eye disc expressing ciScer\UAS.cDa under the control of Scer\GAL4Act5C.PP enter S phase (they are normally arrested in G1). Expression of ciScer\UAS.cDa under the control of Scer\GAL4C96 promotes S phase in G1-arrested cells in the wing margin in wing discs. Clones in the wing disc expressing ciScer\UAS.cDa under the control of Scer\GAL4Act5C.PP are significantly larger than control clones, covering 143% of the area covered by control clones. Cell proliferation rates in the clone are accelerated, but individual cell size and overall cell cycle phasing are unaffected.

Cells in the far anterior compartment of the wing imaginal disc expressing ciScer\UAS.cDa under the control of Scer\GAL4Act5C.PP have wiggly borders to their neighbours. In contrast, cells in the posterior compartment of the wing imaginal disc expressing ciScer\UAS.cDa under the control of Scer\GAL4Act5C.PP sort out from wild-type posterior cells. Clones of posterior origin in the wing disc which are expressing ciScer\UAS.cDa under the control of Scer\GAL4Act5C.PP take up positions normally occupied only by anterior cells and define straight borders to wild-type posterior cells. Anterior clones are strictly confined to anterior territory.

The biomb-3198 mutation prevents the misplacing of ciScer\UAS.cDa, Scer\GAL4hh-Gal4 clones that are generated along the A/P boundary into the A territory.

Co-expression of Rbf280.Scer\UAS does not block the ability of ciScer\UAS.cDa (expressed under the control of Scer\GAL4Act5C.PP) to induce S phase in clones in the morphogenetic furrow. Co-expression of Rbf280.Scer\UAS does not block the ability of ciScer\UAS.cDa (expressed under the control of Scer\GAL4C96) to induce S phase in cells in the wing margin. Co-expression of dapScer\UAS.cLa blocks the ability of ciScer\UAS.cDa (expressed under the control of Scer\GAL4Act5C.PP) to induce S phase in clones in the morphogenetic furrow. Expression of ciScer\UAS.cDa under the control of Scer\GAL4Act5C.PP does not significantly increase the size of clones induced in the wing disc in a Cdk43 mutant background.
