An Hsp70 promoter drives expression of a nkd cDNA fragment (that lacks 5' and 3' untranslated regions) that is tagged at the carboxy-terminal end with Tag:MYC. An Hsp70 3' untranslated region is also present.

Heterozygous wgl-12 embryos kept at the restrictive temperature are patterned normally. Heterozygous wgl-12 embryos also expressing nkdhs.T:Hsap\MYC (using heat shock) secrete cuticles with denticle belt fusions and an excess of type 5 denticles. Homozygous wgl-12 embryos kept at the restrictive temperature have a uniform lawn of type 5 denticles and are unaffected by expression of nkdhs.T:Hsap\MYC.

Comments

Rescues the nkd3 cuticle phenotype when expressed using heat shock. The strongest rescue is seen when 2-4 hour embryos are heat shocked for 15 minutes at 37oC.
