Description

A Tag:MYC epitope has been incorporated immediately following a heterologous signal sequence used to replace the endogenous signal sequence. This epitope is present at the extracellular N-terminus of the receptor coding region. Expression is driven by Mhc promoter sequences.

In the adult prothorax and neck, Myc-tagged GluRIIA protein is observed at the type I boutons of motor neurons innervating ventral cervical muscle 25 and prothoracic sternal adductor muscle 33 from the first day of adulthood, and ventral cervical muscle 26 from the third day of adulthood. GluRIIA protein is also observed at the type I boutons of all adult abdominal muscles. GluRIIA protein is not detected in Type II boutons.

The larval locomotion deficit (reduced crawling ability) characteristic for Msp300ΔKASH mutant third instar larvae is significantly rescued by combination with GluRIIAMhc.T:Hsap\MYC but only in about 60% of the larvae, the remaining 40% show no improvement of the locomotion phenotype.
