kuzk01405 germ line clones exhibit duplication of the RP2 neurons.

Homozygotes have severely reduced wings and halteres, small nota, shortened legs with fused tarsal segments and rough eyes. The flies have gross alterations in the pattern of sensory organs, including the appearance of abnormal tufts of bristles or other sensory organs in locations where only one sensory organ or naked cuticle is present in wild-type flies. Cuticle lacking both microchaetae and macrochaetae is also seen. Homozygous clones induced in the dorsal part of the wing at 60-84 hours after egg laying give rise to thickening of the wing veins and nicks at the wing margin. Within the clone a cluster of campaniform sensilla develops at the position of one of the twin sensilla of the margin. kuz1405Rev4/kuzk01405 flies show scalloping of the wings. Homozygous larvae have imaginal discs of reduced size. The pupal notum contains lawns of neurons where only one or a few neurons are present in wild-type flies.

The scalloping of the wing margin seen in SerD/+ heterozygotes is enhanced if the flies also carry kuz1405Rev4/kuzk01405. The neurogenic phenotype of kuzk01405 flies is partially rescued by E(spl)Scer\UAS.cSa expressed under the control of Scer\GAL4C-765; many single sensory organs, rather than tufts of sensory organs, develop on the notum. The sensory organ but not the wing vein phenotypes of H2 heterozygotes and homozygotes are suppressed by kuzk01405. The neurogenic phenotype and reduced wing size of kuzk01405 homozygotes is suppressed by H2. kuzk01405 is partially rescued by Nint.hs at 25oC; kuzk01405 flies carrying Nint.hs have a normal-sized notum and eyes, and their legs, wings and halteres are substantially larger than those of kuzk01405 single mutant flies.

Comments

The development of wings, halteres, thorax, legs and eyes in homozygotes is rescued by kuzScer\UAS.cFa expressed under the control of Scer\GAL4C-765. The pattern of sensory organs is restored almost to normal in these flies, although several notum microchaetae are missing and extra stout bristles (some with abnormal morphology) are seen on the anterior wing margin.

Comments
Comments

Precise excision of the P{lacW} element results in reversion to wild-type.
