    Rca12 mutant animals fail to execute mitosis after the 16th cell cycle, and subsequently embryos have fewer cells compared to wild-type.

    Rca12 mutant clones generated in the wing imaginal disc have a severe proliferation disadvantage, and are much reduced in size compared to wild-type clones. Mutant cells undergo endoreplication of the DNA, and have an increased nuclear size.

    Rca12/Rca13 embryos exhibit a failure of cell division in the epidermis at mitotic cycle 16: No mitotic spindles are seen at the time when wild-type cells would normally undergo this division, and the DNA is decondensed, suggesting arrest in interphase. Rca12 homozygotes also exhibit cycle 16 cell cycle arrest. Somatic clones of Rca12 in the wing disc are smaller than their corresponding wild-type twinspots. The presence of twinspots in the absence of a corresponding Rca12 mutant clone suggests that mutant cells are sometimes eliminated from the disc.

    Rca12/Rca13 transheterozygous embryos arrest in cell cycle 16.

    CycAScer\UAS.T:Ivir\HA1; Scer\GAL4prd.RG1 suppresses the mitotic block due to Rca12/Rca12, in alternate segments. CycEScer\UAS.cLa; Scer\GAL4prd.RG1 suppresses the mitotic block due to Rca12/Rca12, in alternate segments. The embryonic cycle 16 mitotic block due to Rca12/Rca12 is suppressed by Df(1)bi-D3/Df(1)bi-D3. Cell cycle arrest in CycAC8LR1/CycAC8LR1; Rca12/Rca12 embryos, occurs earlier (cycle 15) than in either single mutant (both arrest at cycle 16).

Clones are increased in size compared to Rca12 mutant clones, but are only a third of the size of wild-type clones. Rarely, endoreplication is observed in mutant clones.

    (with Rca13) Partially rescues: { Rca1<up>Scer\UAS. T:Ivir \HA1</up>, Scer\GAL4prd.RG1 } In Rca12/Rca13; Rca1Scer\UAS.T:Ivir\HA1/Scer\GAL4prd.RG1 embryos, mitosis 16 is rescued in alternating segments resulting in almost twice as many cells in mutant segments as in rescued ones.
