Amino acid change:

W246term | hd-PA; W246term | hd-PB

Reported amino acid change:

W246term

Comment:

G to A nucleotide change at the second or third position of the wild type Trp codon leads to a nonsense mutation (exact site of mutation unspecified). The mutation was annotated at the second base of the codon.

Imaginal discs of hdFf/Df(3R)3-4 transheterozygous larvae have cell proliferation defects.

Clones of homozygous hdFf follicle cells arrest during G2 and early M phases of the cell cycle without evidence of chromosome bridging.

hdFf/Df(3R)3-4 mutants have small brains that display severely reduced levels of DNA synthesis compared to wild type. They also have no identifiable imaginal discs and small, under-replicated polytene chromosomes in the salivary glands. hdFf mutant clones generated in the wing disc are significantly smaller both in terms of size and number of cells than wild-type twin spots.

FACS analysis shows that a higher proportion of hdFf cells from wing disc clones have a 2C or intermediate 2C-4C content than wild-type cells. BrdU incorporation shows that DNA replication is reduced within the hdFf clones. Additionally, cells from the hdFf wing disc mutant clones show a higher proportion of repair foci than wild-type twin spots.

Early pupal lethal.
