Description

Stop codons introduced at codon positions 432 and 433, results in deletion of the last 15 amino acids of the C terminus.

Stop codons introduced in the coding region in codon positions 432 and 433, by two T for G substitutions.

Males carrying one or two copies of βTub85D432 in a wild-type background are fertile. Males carrying four copies of βTub85D432 in a wild-type background are fertile, but have reduced fecundity and a reduced amount of motile sperm compared to wild type. Males carrying one or two copies of βTub85D432 in a βTub85Dn/βTub85Dn background are sterile and axonemes are not produced. Males carrying one copy of βTub85D432 in a βTub85Dn/+ background are sterile.

In the absence of wild type βTub85D, βTub85D432 can support the assembly and function of many classes of microtubules but causes intrinsic defects of differing severity in meiosis, sperm head shaping and axoneme morphogenesis.

Spermatids have normal early post-meiotic morphology. However meiotic divisions are often flawed, due to a failure in cytokinesis. Differing size of nuclei indicate non-disjunction. Spermatid heads remain unshaped. Spermatid elongation proceeds, at least to some degree, though ultrastructural examination reveals that the axoneme is abnormal.

Males carrying one copy of αTub84B+t4.5 and one copy of βTub85D432 in an otherwise wild-type background are sterile. Males carrying one copy of αTub85EβTub85D.PH and one copy of βTub85D432 in an otherwise wild-type background are sterile. Df(3R)Scx4/+ ; βTub85Dn/+ males carrying one copy of βTub85D432 are fertile. Males carrying one copy of βTub56DβTub85D.PR and one copy of βTub85D432 in a βTub85Dn/+ background are fertile, but have reduced fecundity and a reduced amount of motile sperm compared to wild type. Males carrying one copy of βTub60D::βTub85DβTub85D.3'UTR and one copy of βTub85D432 in a βTub85Dn/+ background are sterile and have abnormal axonemes.

Protein product (B2ΔC) can be assembled into ultrastructurally normal looking microtubules in vivo. Phenotypes of males with 1 to 4 copies of this allele are identical.

Encodes the protein product B2ΔC.
