Description

Nucleotide substitution: A104T. Amino acid replacement: K35M.

An amino acid replacement - single substitution in stnA.

Lesion may map to the stnA rather than the stnB transcription unit.

Contains the InsOR20B 2.4kb insertion polymorphism present in Oregon-R but not Canton-S.

stnA7 larvae show approximately threefold enhancement in the frequency of spontaneous miniature events at the neuromuscular junction compare to wild-type. Postsynaptic responses in the muscle evoked by motor nerve stimulation are not substantially altered compared to wild-type. The fractional area occupied by vesicles is not significantly different from wild-type in stnA15 boutons at the neuromuscular junction, although there is a trend towards wider synaptic vesicle distribution than in wild-type. The synaptic vesicles are indistinguishable from wild-type and synaptic vesicle density is normal. The number of boutons per arbor is slightly, but significantly, increased compared to wild-type.

At 22oC flies walk slowly, occasionally fall over, flutter wings, can climb and fly. At 29oC flies fall to the bottom of the vial, are paralyzed, though incompletely, as their legs kick as they lie on their backs. Mutant phenotype is reversible by shifting back to low temperature. Heterozygotes with Df(1)30A emerge late, become mired in the medium, and show low relative viability. ERG phenotype shows increased amplitude of off-transient. Flies heterozygous for a ts and a lethal allele of stnA show ERGs with off transient amplitudes between those for stnA7 and stnA15. Flies heterozygous for stnA15 and a ts allele show ERG off transients significantly lower than for the control class, and lower than for the heterozygotes between ts and lethal alleles of stnA.

Unusual jumping reaction in response to a light off stimulus and unable to coordinate movements at the restrictive temperature.

Homozygous flies can walk at the restrictive temperature, but do so with difficulty.

Severe behavioral debilitation is quickly induced by shift of adult stnA7 from 25oC to 29oC; abnormalities at high temperature include uncoordinated leg and wing movements, with complete paralysis never occurring (e.g., legs still move); stnA7 is more nearly normal at low temperatures and can even walk with difficulty at 29oC; at permissive temperatures.

The viability of sytD27/sytAD4 flies (rescued to adulthood by sytelav.PD) is significantly reduced in combination with stnA7/Y. The few surviving males have an extreme behavioural phenotype and are very sedentary and severely uncoordinated (they are more uncoordinated than stnA+/Y ; sytD27/sytAD4 flies rescued to adulthood by sytelav.PD). They die within 48 hours of eclosion.

Allele specific synergistic interaction with mutations at the shi and dnc loci.

The lethality of shi1, stnA7 double mutants is complete. The Sh7, stnA7, parats1, stnA7 and rut-,stnA7 double mutants are no more inviable than stnA7 alone. Su(stn)1 suppresses the lethal interaction of dnc1, stnA7.

Fails to complement
Comments

Fails to fully complement the reduced excitatory junctional potential amplitude of stnA15.

stnA15 entirely complements the uncoordinated phenotype of stnA7, though stnA7 does not complement the stnA15 stress-sensitive behavior.

Discoverer

Putative cluster-mate of stnA6; both stnA6 and stnA7 were isolated from the same strain and have the same nucleotide substitution.
