Comments

Approximately 60.5 kb deletion which removes Ilp1 to Ilp5 and nearby genes resulting from FLP-catalyzed recombination using the FRT-containing transposons P{XP}CG8177d02657 and PBac{WH}CG43897f05433.

Central nervous system neuroblasts of fed Df(3L)Ilp1-5 second instar larvae show a strong reduction in EdU incorporation compared to controls. Despite this delay in neuroblast reactivation, it eventually begins in the normal pattern (albeit heterochronically) in larvae with third instar morphology.

Homozygous Df(3L)Ilp1-5 flies are viable and fertile, but they compete poorly with wild-type or heterozygous animals. Homozygous Df(3L)Ilp1-5 animals display reduced fertility compared to wild-types. Their total lifetime egg production and mean maximal number of eggs laid per day are decreased to about 10% of the level of control animals. Viability of Df(3L)Ilp1-5 homozygotes is decreased compared to wild-types. Development of Df(3L)Ilp1-5 homozygotes to the pupal stage is decreased approximately 45% of the that of controls animals and development to adult stage is decreased to approximately 41% of that of control animals.

Df(3L)Ilp1-5 homozygous adults are small with decreased body mass and are developmentally delayed. The developmental time to reach the wandering third instar stage is lengthened to more than 9 days, compared to 5 days for controls, and the time to reach pupation is also extended, but the time from pupation to eclosion is not greatly affected. None of these phenotypic defects are observed in Df(3L)Ilp1-5 heterozygotes.

Wing area of homozygous Df(3L)Ilp1-5 flies is smaller than that of wild-type controls due decreases in both cell size and cell number. Cell size in the fat body of homozygous Df(3L)Ilp1-5 animals is decreased compared to controls.

In Df(3L)Ilp1-5 homozygotes, male genital arch posterior lobes fail to scale proportionately with the smaller body and smaller wing size.

Df(3L)Ilp1-5 homozygotes show a decrease in normalised whole body triglyceride levels compared to wild-type animals. Actively feeding homozygous Df(3L)Ilp1-5 larvae, unlike wild-type controls, exhibit starvation response in the fat body.

Homozygous Df(3L)Ilp1-5 larvae are less active than wild-type animals and the mouth hook contraction rate appears lower than that of controls.

The overall metabolic rate of Df(3L)Ilp1-5 homozygotes is reduced compared to wild-type animals. Circulating sugar levels are elevated in the hemolymph of homozygous Df(3L)Ilp1-5 larvae and adults.
