Description

Transgenic construct used in a study to test the ability of insulator or boundary elements to block repression of a target gene by a Polycomb response element (PRE). Order of sequences (from P3' to P5') is: y+mDint2 marker (plus orientation) , a 436bp BalI-BstXI fragment from the gypsy\su(Hw)BR (containing 12 su(Hw) protein binding sites), a w+mC marker (minus orientation), a 661bp NdeI-PstI fragment containing the core activity of the major Ubx Polycomb response element (PRE, located approximately 24kb upstream of the Ubx promoter), a 436bp BalI-BstXI fragment from the gypsy\su(Hw)BR (containing 12 su(Hw) protein binding sites).
