M{MlwB} is an enhancer trap construct that contains at its 5' end the weak P\T promoter fused upstream of sequence encoding the lacZ reporter enzyme (the lacZ sequence is fused in-frame to the second exon of the P\T coding sequence). In addition, the endogenous (weak) Dmau\mariner\T promoter is also present approximately 700bp upstream of the enhancer trap sequence and could potentially effect lacZ expression.
Construct: Fragment of Mos1 up to nucleotide 787, including the Dmau\mariner 5' terminal repeat.
Dmau\mariner\T coding region interrupted at position 789 by introduction of w+mW.hs.
Insertion of foreign DNA, including w+mW.hs, in the SacI site at position 789 of the Mos1 transposable element.
Basal pattern of stripes, restricted to epidermal clusters of cells. Expression localized to nucleus.
Expression pattern dependent upon position of insertion in genome. Basal pattern observed in approximately 30% of transformants: in extended germ band embryos, weak expression in dorsal epidermis, stronger in alternate segments.
