Description

7 copies of a 30bp EcR element unit from the Hsp27 promoter (23bp element and 7 flanking bp) fused upstream of an Adh TATA box located within a BglII-ScaI fragment of the Adh distal promoter (-34 to +53) fused upstream of lacZ coding region.

lacZ expression is driven by seven copies of the EcR response element.

The reporter was used to monitor EcR/usp activity in 0-, 5- and 10-day adults. Activity was observed in the Malpighian tubules and accessory glands at all times. Activity was rarely observed after day 0 in the CNS, abdominal cuticle fat, and seminal vesicles. Activity in adult oenocytes increased after day 0. Activity was observed within the first few hours after eclosion in testis but was absent by day 5. Activity in the ejaculatory bulb and duct varied over the time period. Activity was also observed in follicle cells and gut.

In third instar larval eye disc, expression restricted to a zone in and slightly anterior to the morphogenetic furrow; first observed when the morphogenetic furrow has produced 10-12 columns of ommatidia; travels with the furrow until the end of third instar, when it becomes ubiquitous.

Comments

report comment: P{EcRE.lacZ} carries a reporter gene consisting of 7 copies of a 30bp sequence from Hsp27, which includes the 23-bp EcRE region; these are followed by the Adh distal promoter; followed by the lacZ gene.
