The following information accompanied stocks donated to the Bloomington Stock Center by Douglas Kim, Genetically-Encoded Neuronal Indicator and Effector (GENIE) Project, Janelia Research Campus. Constructs expressing improved fluorescent calcium-sensor proteins were made in a vector similar to pJFRC13 (see http://flybase.org/reports/FBrf0218136.html) with 20 copies of UAS, an intervening sequence from the Drosophila myosin heavy chain gene, a nuclear export signal and the Autographa californica nucleopolyhedrovirus p10 gene transcription terminator. PBac{20XUAS-IVS-NES-jRCaMP1a-p10} expresses a bright red variant of RCaMP with amino acid substitutions T62C, Y112S and L298T relative to the RCaMP1h progenitor (http://flybase.org/reports/FBrf0221174.html). This variant RCaMP provides an improved signal-to-noise ratio and may be used in combination with Channelrhodopsin-2. PBac{20XUAS-IVS-NES-jRCaMP1b-p10} expresses a bright red variant of RCaMP with amino acid substitutions T46W, I60N, L326M and R368G relative to the RCaMP1h progenitor (http://flybase.org/reports/FBrf0221174.html). This variant RCaMP provides an improved signal-to-noise ratio, faster decay time and may be used in combination with Channelrhodopsin-2. PBac{20XUAS-IVS-NES-jRGECO1a-p10} expresses a red variant of R-GECO1.0 with amino acid substitutions Q306D and M339F relative to the R-GECO1.0 progenitor (Zhao et al. (2011) Science 333: 1888-1891; Li et al. (2013) PNAS 110: 12084-12089). This variant provides an improved signal-to-noise ratio. PBac{20XUAS-IVS-NES-jRGECO1b-p10} expresses a red variant of R-GECO1.0 with amino acid substitutions I109K, R131C and L321C relative to the R-GECO1.0 progenitor (Zhao et al. (2011) Science 333: 1888-1991; Li et al. (2013) PNAS 110: 12084-12089). This variant provides an improved signal-to-noise ratio and improved dynamic range.
