Under 12h:12h light:dark cycle conditions, the conditional expression of nurUAS.Tag:FLAG,Tag:HA only during adulthood under the combined control of Scer\GAL4elav.Switch.PO and RU486 feeding results in a significant increase in sleep duration during both daytime and nighttime periods in both males and females, as compared to controls; during daytime, there are significantly more sleep bouts but no change in bout duration; during nighttime, there is a significant increase in bout duration despite of significantly fewer sleep bouts; although mid-nighttime mechanical stimulation awakes nearly all control flies, it only awakes some of nurUAS.Tag:FLAG,Tag:HA-expressing flies; the few aroused flies are sluggish relative to controls but this does not seem related to overall locomotor impairment, as it even leads to significant increase in speed movement 30min after lights on/lights off, as compared to controls.
The same conditional adult expression leads to increased survival to infection with S. pneumoniae (associated with increased daytime sleep duration) and S. marcescens (associated with increased daytime sleep duration and with lower bacteria load). In the absence of infection, it does not affect lifespan, does not affect food intake and induces only mild circadian defects (i.e. the circadian period is slightly but significantly shorter under free-running/constant darkness conditions but behavioral rhythms/FFT are clearly defined), as compared to controls.
The conditional expression of nurUAS.Tag:FLAG,Tag:HA only during adulthood under the combined control of Scer\GAL4Switch1.106 and RU486 feeding does not affect sleep duration in the absence of infection, nor does it affect survival or bacterial load upon infection by S. marcescens.

The increased sleep induced by conditional adulthood-only expression of nurUAS.Tag:FLAG,Tag:HA under the combined control of Scer\GAL4elav.Switch.PO and RU486 feeding is not suppressed by silencing Ecol\lexAGMR23E10-expressing neurons through Hsap\KCNJ2lexAop.EGFP expression (Ecol\lexAGMR23E10>Hsap\KCNJ2lexAop.EGFP).
