UASt regulates expression of the Hsap\LAMP2 cDNA (clone H05D091G15. (The sequence upstream of the ATG initiation codon was modified to match the Drosophila translation start consensus sequence.)
Individuals expressing Hsap\LAMP2UAS.cIa under the control of Scer\GAL4elav-C155 develop normally to adults and do not show apparent behavioral defects. Adults show longer lifespan upon starvation or paraquat-induced oxidative stress, and increased climbing ability but no change in lifespan under control conditions. These individuals show significantly increased numbers of KFERQ-positive puncta (marker of late endosomes and lysosomes) in the adult brain under fed conditions, but not under starvation, nor in the larval fat body under fed or starvation conditions.
