The VT019059 enhancer fragment followed by the Drosophila synthetic core promoter (DSCP) is fused upstream of the GAL4(DBD)::Zip- hemidriver (component of the 'Split GAL4' system, contains the GAL4 DNA-binding domain).

Thermogenetic activation (using TrpA1UAS.cvPa at 30[o]C) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.VT019059 and Hsap\RELAAD.VT008469, or the split-GAL4 pair of Scer\GAL4DBD.VT019059 and Hsap\RELAAD.VT050660, results in a small, significant increase in the total distance, but not the bout distance, of backwards walking, with a significant decrease in forwards walking, compared to controls.
Inhibition (using Ctet\TeTxLCTNT.UAS) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.VT019059 and Hsap\RELAAD.VT008469, or the split-GAL4 pair of Scer\GAL4DBD.VT019059 and Hsap\RELAAD.VT050660, does not significantly affect backing-up behavior when adult flies reach a narrow dead end, compared to controls; there is no significant effect on walking direction in unimpeded conditions, compared to controls.
