UAS regulatory sequences drive expression of an inverted repeat of a 580bp fragment of dnc transcript D (nucleotides 1617-2197) which targets five exons (exons 10-14 of transcript D) that are common to all dnc isoforms.

Adult flies expressing dncdsRNA.Scer\UAS.WIZ under the control of Scer\GAL4elav.PU show significant decrease in the amnestic cooling resistant memory component at 3 hr post-training in an olfactory memory assay compared to controls.

Expression of dncdsRNA.UAS.WIZ under the control of Scer\GAL4c739 in adults significantly reduces the anaesthesia-resistant component of short term (5 min) memory, compared to controls; expression under the control of Scer\GAL4NP0021 has no effect on the anaesthesia-resistant component of medium term (3 h) aversive olfactory memory, compared to controls; expression under the control of Scer\GAL44-59 significantly reduces long term (24 h after massed training) aversive olfactory memory, compared to controls.

Expression of dncdsRNA.Scer\UAS.WIZ under the control of Scer\GAL4elav.PU results in defective short term memory, 3 min after olfactory conditioning, and defective anaethesia-resistant memory, but not defective anaesthesia-sensitive memory, when cold-amnestic treatment is applied 30 min before testing 3 h odor memory, as compared to controls.

Expression of dncdsRNA.Scer\UAS.WIZ under the control of Scer\GAL4GH298 or Scer\GAL4NP1227, but not Scer\GAL4Orco.PU, Scer\GAL4Mef2.247 or Scer\GAL4NP0225, significantly decreases short term memory performance 3 min after olfactory conditioning.

Expression of dncdsRNA.Scer\UAS.WIZ under the control of Scer\GAL4GH298, but not Scer\GAL4Mef2.247, results in defective anaethesia-resistant memory, but not defective anaesthesia-sensitive memory, when cold-amnestic treatment is applied 30 min before testing 3 h odor memory, as compared to controls.
