UASt regulatory sequences drive expression of Hsap\TARDBP carrying the amino acid replacement G287S (a causal ALS mutation).
Expression of Hsap\TARDBPG287S.Scer\UAS.cVBa driven by Scer\GAL4GMR.PU does not induce a rough eye phenotype and expression driven by Scer\GAL4nSyb.PU or Scer\GAL4elav-C155 does not cause wing expansion defects. With expression driven by Scer\GAL4nSyb.PU, there is no significant or progressive loss of Kenyon cells, or significant changes in the length or thickness of the alpha lobe in 3 or 50 day old flies compared to controls; there are no significant changes in the length of the beta lobe in 3 or 50 day old flies, though there is a significant decrease in the thickness of the beta lobe of 50 (but not 3) day old flies. Expression driven by Scer\GAL4CCAP.PP results in predominantly nuclear localization of the protein, but with a shift to the cytoplasm in around 12-14% of bursicon neurons in the ventral nerve cord.

However, the median lifespan of surviving TBPHΔ142/TBPHΔ23 flies with expression of Hsap\TARDBPG287S.Scer\UAS.cVBa is shorter than in controls.
