UASt regulatory sequences drive expression of the complete open reading frame of the Hsap\NUMB 'PTB[S]PRR[L]' alternative isoform (isoform 3), which contains the short version of the PTB domain and the long version of the PRR domain (contains a 48 amino acid residue insertion relative to the short form).
Compared to control clones in third instar larval brains, clones with expression of Hsap\NUMBScer\UAS.PTBS-PRRL have an increased number of neuroepithelial cells and neuroblasts contain more neurons.
Overexpression of Hsap\NUMBScer\UAS.PTBL-PRRS (or Hsap\NUMBScer\UAS.PTBS-PRRS) leads to dramatic wing margin balding (much more severe than Hsap\NUMBScer\UAS.PTBS-PRRL), when driven by Scer\GAL4bbg-C96, compared to controls; this represents a transformation toward neuronal fate.
Scer\GAL4Tub.PU/Hsap\NUMBUAS.PTBS-PRRL is a suppressor of abnormal neuroanatomy | somatic clone | third instar larval stage phenotype of numb15
Scer\GAL4Tub.PU/Hsap\NUMBUAS.PTBS-PRRL is a suppressor of decreased cell number | somatic clone | third instar larval stage phenotype of numb15
Hsap\NUMBUAS.PTBS-PRRL is a non-suppressor of decreased cell number | somatic clone | third instar larval stage phenotype of NICN.UAS
Scer\GAL4Tub.PU/Hsap\NUMBUAS.PTBS-PRRL is a suppressor of neuron | somatic clone | third instar larval stage phenotype of numb15
Hsap\NUMBUAS.PTBS-PRRL is a non-suppressor of neuron | somatic clone | third instar larval stage phenotype of NICN.UAS
Co-expression of Hsap\NUMBScer\UAS.PTBS-PRRL increases the number of neuroepithelial cells but does not suppress the inhibition of neuronal differentiation seen in clones with expression of NICN.Scer\UAS in third instar larval neuroblasts.
Expression of Hsap\NUMBScer\UAS.PTBS-PRRL driven by Scer\GAL4tub.PU partially suppresses neuronal differentiation defects seen in the outer optic anlage of numb15/numb15 clones in third instar larvae.
