Comment:

Estimated extent of a 3224 bp deletion resulting from the imprecise excision of P{GSV7}GS21694. The excision is reported to include the 5' UTR and 505aa (methods) or 508aa (results) of the CPT2 coding sequence. This mapping assumes 505 codons have been completely removed.

Detailed Description

The majority of CPT2δ3 animals die during the first 3 days after eclosion - very few die during larval or pupal development. Those that survive the immediate post-eclosion period show a severely reduced adult lifespan and are hyper-sensitive to starvation.

Normally fed CPT2δ3 flies are not heaver, but even lighter than controls. Body length is somewhat reduced, but the ratio of weight and size (BMI) is not increased, compared to controls. Motor activity is also unchanged. However, while total body triacylglyceride (TAG) levels are not increased, there is massive accumulation of TAG-filled lipid droplets in the brains and flight muscle in these mutant flies.

Glial-specific (Scer\GAL4repo.PU) expression of CPT2Scer\UAS.cSa reverses brain TAG accumulation and peri-eclosion lethality of CPT2δ3 mutants.

Muscle-specific (Scer\GAL4Mef2.PU), but not fat body (Scer\GAL4cg.PU) or neuronal (Scer\GAL4nSyb.PU), expression of CPT2Scer\UAS.cSa reverses peri-eclosion lethality of CPT2δ3 mutants.

Expression of CPT2Scer\UAS.cSa in cortex glia (Scer\GAL4Akap200-NP2222 or Scer\GAL4nrv2.PU) or astrocyte-like glia (Scer\GAL4Eaat1.PU), but not in ensheathing glia (Scer\GAL4NP6520), reverses peri-eclosion lethality of CPT2δ3 mutants.
