UAS regulatory sequences drive expression of the spGFP11 split fluorescent protein fragment fused to a membrane targeting sequence (Tag:M(hCD4)). This construct is designed for use in a GRASP (GFP Reconstitution Across Synaptic Partners) assay, in which complementary split fluorescent protein fragments expressed on the extracellular membrane of different neurons can reconstitute a functional fluorescent protein at points of contact between the neurons.
GRASP flies co-expressing nSybEcol\lexAop.T:Avic\GFP-spGFP1-10 (under the control of Ecol\lexAIr93a.PU) and Avic\GFPspGFP11.Scer\UAS.T:Hsap\CD4 (under the control of Scer\GAL4VT040053) do not show any changes in temperature preference compared with controls.
