The RMCE cassette of the Mi{MIC}dlg1MI06353 insertion has been replaced with a EGFP-FlAsH-StrepII-TEV-3xFlag multi-tag cassette. The insertion is in a coding intron of dlg1 and is predicted to protein trap the gene.
dlg1MI06353-GFSTF.0 flies in which the level of dlg1 protein has been reduced by expressing slmbScer\UAS.T:Zzzz\vhhGFP4 (which targets GFP-tagged proteins for degradation) under the control of Scer\GAL4tub.PU develop normally and eclose without obvious phenotype at 18[o]C, but show embryonic lethality when raised continuously at 28[o]C. Animals which are shifted to 28[o]C during the first or second larval instar show hyperproliferation of the imaginal disc and brain as third instar larvae and show a mild decrease in neuromuscular junction growth (in terms of bouton number and synapse complexity). Animals which are shifted to 28[o]C during the first larval instar show defects in cell morphology and organisation in the larval midgut.
