Description

UAS regulatory sequences drive expression of a Zzzz\CAG-repeat encoding 90 alanines. The repeats are flanked by seven amino acids on the N-terminal side and four on the C-terminal, and the Tag:MYC and Tag:FLAG epitope tags are located downstream.

Expression of one copy of Zzzz\Poly-Ala90.Scer\UAS.T:Hsap\MYC,T:Zzzz\FLAG under the control of Scer\GAL4GMR.PFa has no effect on the external eye, although there is a mild defect in the organisation of the underlying structure, with some alterations in photoreceptor number, as well as a mild disorganisation of pigment granules surrounding the ommatidial units. No aggregates are observed in the eye. Expression of two copies of the transgene results in a rough eye phenotype with a decrease in size and some loss of pigmentation. Variable-sized aggregates are seen in the eye. The structure of the lamina is also disrupted.

Flies expressing Zzzz\Poly-Ala90.Scer\UAS.T:Hsap\MYC,T:Zzzz\FLAG under the control of Scer\GAL4da.G32 are viable. Zzzz\Poly-Ala is seen in both the nucleus and cytoplasm, but aggregates form predominantly in the cytoplasm in third instar larvae. Adult flies exhibit a severe defect on the patterning of the abdomen, where the tergites bands frequently appear split at the midline.

Flies expressing Zzzz\Poly-Ala90.Scer\UAS.T:Hsap\MYC,T:Zzzz\FLAG under the control of Scer\GAL4elav-C155 show climbing defects.

Brains from adult flies expressing Zzzz\Poly-Ala90.Scer\UAS.T:Hsap\MYC,T:Zzzz\FLAG under the control of Scer\GAL4elav.PLu show no evidence of aggregates.
