21.8% of pupae expressing Aats-serdsRNA.Scer\UAS.WIZ under the control of Scer\GAL4Act5C.PU (in the presence of Dcr-2Scer\UAS.cDa to enhance RNAi efficiency) hatch successfully at 25[o]C. No adults hatch at 29[o]C.
Mitochondrial ultrastructure is abnormal in larvae expressing Aats-serdsRNA.Scer\UAS.WIZ under the control of Scer\GAL4Act5C.PU (in the presence of Dcr-2Scer\UAS.cDa) at 29[o]C. The matrices are swollen and electron pale, there is a loss of mitochondrial cristae and the mitochondria are significantly enlarged.
Lactate concentration is increased compared to wild type in larvae expressing Aats-serdsRNA.Scer\UAS.WIZ under the control of Scer\GAL4Act5C.PU (in the presence of Dcr-2Scer\UAS.cDa). Glycogen concentration is decreased.
Animals expressing Aats-serdsRNA.Scer\UAS.WIZ under the control of Scer\GAL4nub-AC-62 (in the presence of Dcr-2Scer\UAS.cDa) show tissue damage in the wing, with a penetrance of 32.3% at 25[o]C and 97.7% at 29[o]C. Relative mtDNA copy number is increased compared to wild type.
Partial or total loss of the anterior crossvein is seen in 4% of flies expressing Aats-serdsRNA.Scer\UAS.WIZ under the control of Scer\GAL4ptc-559.1 (in the presence of Dcr-2Scer\UAS.cDa) at 29[o]C. The L3-L4 intervein region is significantly reduced in area compared to wild type, with the reduction being due to a decrease in cell number and not in cell size.
Flies expressing Aats-serdsRNA.Scer\UAS.WIZ under the control of Scer\GAL4repo.PU (in the presence of Dcr-2Scer\UAS.cDa) have an altered survival curve compared to controls, with a reduction in half-life from 32 to 21 days at 29[o]C.
Adults expressing Aats-serdsRNA.Scer\UAS.WIZ under the control of Scer\GAL4Mef2.PU (in the presence of Dcr-2Scer\UAS.cDa) have reduced climbing ability compared to controls.
