PIP4K29 individuals show delayed pupariation under normal diet and earlier pupariation under high sugar diet, as compared to controls.

The photoreceptors of PIP4K29 homozygous late pupae (i.e. at 90% of pupal development) reared in light conditions exhibit significant increases in the number and the size of endosome-like vesicles, a significant expansion of multi-vesicular bodies in the cell bodies, and upregulation of early endosomes (assessed by EEA1 or Rab5 Western blot and/or immuno-staining), but do not exhibit obvious defects in the distribution of endoplasmic reticulum (identified by KDEL immuno-staining) and of the Golgi apparatus (identified by GM130 immuno-staining), as compared to controls; these mutants reared in light conditions do not exhibit any obvious defects in the structure of rhabdomeres, or in the levels of autophagosomes in head extracts (assessed by Atg8a protein levels by Western blot or by puncta of fluorescent human LC3 protein), as compared to controls. The haemocytes of PIP4K29 homozygous third instar larvae display a significant increase in clathrin-dependent endocytosis, as compared to controls.

Light responses are normal in PIP4K29 photoreceptors.

Homozygous adults have phosphatidylinositol 5-phosphate levels double that of wild-type controls, while the levels of phosphatidylinositol 4-phosphate and phosphatidylinositol (4,5)-bisphosphate are not reduced compared to wild type.

Homozygous larvae show developmental defects. Although they have the same body weight as controls immediately after hatching, they weigh less than wild-type controls at multiple points during development, with the maximum difference being at 60 hours after hatching at 22[o]C. The mutant larvae show a delay in pupariation compared to wild type. Salivary gland cells are reduced in size in the mutant larvae compared to wild type.

PIP4K29 third instar larvae expressing RhebUAS.cPa under the control of Scer\GAL4AB1 show significantly bigger salivary gland cells, as compared to PIP4K29 single mutants.

The expansion in endosome-like vesicles and multivesicular bodies observed in photoreceptors of PIP4K29 homozygous late pupae reared in light conditions are suppressed by the expression of Rab5S43N.Scer\UAS.P\T.T:Avic\GFP-YFP, but not RhebScer\UAS.cPa, under the control of Scer\GAL4ninaE.PU.
