Expression of dsxUAS.female under the control of Scer\GAL4insc-Mz1407 from mid second instar stage, in combination with a Gal80[ts] transgene, results in a reduced number of neuroblasts in the thoracic segments of the late third instar male larval ventral nerve cord, compared to controls; expression in females from early first instar stage does not prevent female-specific loss of larval terminal neuroblasts, compared to controls.

Under control conditions, the adulthood-only expression of dsxUAS.female under the control of Scer\GAL4esg-NP7397 (together with Gal80[ts], for the temporal regulation of expression, and Dicer-2, for efficient RNAi) does not significantly affect the number of progenitor cells (intestinal stem cells/enteroblasts) or the mitotic index in the female or male adult posterior midgut, as compared to controls.

Expression of dsxF.Scer\UAS under the control of Scer\GAL4dsx.KI results in single-X males with female-like abdominal pigmentation, genitalia and no sex combs.

Female flies expressing both dsxM.Scer\UAS and dsxF.Scer\UAS under the control of Scer\GAL4dsx.KI show a significant increase in neuron numbers in dsx-pC1, dsx-pC2 and dsx-SN (but not dsx-pC3) clusters in the brain and in all of the VNC clusters compared with wild-type females with a concomitant increase in projections and formation of contralateral connections. However, the number of dsx-pC1 and dsx-pC2 neurons is still significantly lower than in wild-type males.

Expression of Abd-BGD4680 and dsxUAS.female, in combination with a Dicer-2 transgene to enhance RNAi efficiency, under the control of Scer\GAL4insc-Mz1407 from early first instar stage, in combination with a Gal80[ts] transgene, prevents female-specific loss of larval terminal neuroblasts, compared to controls; there is also a significant decrease in the number of thoracic neuroblasts in the female, compared to controls.

Under control conditions, the adulthood-only co-expression of dsxUAS.female and traJF03132 under the control of Scer\GAL4esg-NP7397 (together with Gal80[ts], for the temporal regulation of expression, and Dicer-2, for efficient RNAi) does not significantly affect the number of progenitor cells (intestinal stem cells/enteroblasts) or the mitotic index in the female adult posterior midgut, as compared to controls.
