A sub-fragment of the genomic ple sequence present in Scer\GAL4ple.PF is fused upstream of the GAL4 coding region.
ScerGAL4ple.TH-D drives expression in a subset of ple-positive dopaminergic neurons in the adult brain as well as in the thorax and abdomen.
ScerGAL4ple.TH-D' drives expression in a subset of the dopaminergic neurons in the adult brain labelled by ScerGAL4ple.PF : around 3 PPM2 neurons, 6 PPM3 neurons and 10 PPL1 neurons. There was also some non-systematic expression in less than 1 neuron in the PAM and PPM1 clusters.
Activation of neurons expressing any of the specific dopaminergic neuron drivers Scer\GAL4ple.TH-D, Scer\GAL4ple.TH-D-4 or Scer\GAL4ple.TH-F-1 through overexpression of temperature sensitive TrpA1 results in increased sleep loss in both mature (7-8 day) flies compared to controls. Expression under the control of Scer\GAL4ple.TH-D or Scer\GAL4ple.TH-D-4 also results in sleep loss in young (1 day old) flies. The resulting Scer\GAL4ple.TH-D-4>TrpA1 mature flies show a deficit in courtship measures and a reduction in VA1v volume. DA1 and VA1d volumes are unaffected. No difference in sleep amount in young flies is seen when neurons expressing Scer\GAL4ple.TH-F-1 are activated. Activating neurons that express in ventral projections to the antennal lobe (AL) in young flies using Scer\GAL4ple.TH-C results in sleep loss, but the flies do not show courtship deficits or glomerular volumetric changes in mature adulthood.
