Description

UAS regulatory sequences drive expression of a DsRed(Unk) fluorescent protein. A genomic fragment containing the mir-14 pre-miRNA hairpin has been inserted into the DsRed(Unk) 3'UTR.

Overexpression of mir-14Scer\UAS.cLa.T:Disc\RFP driven by Scer\GAL4hh-Gal4 causes curvature of the wing and significantly decreases posterior (P) wing compartment size and the posterior / anterior (A) wing size ratio compared to controls. Overexpression of mir-14Scer\UAS.cLa.T:Disc\RFP by Scer\GAL4sd.PU significantly reduces wing size. Overexpression of mir-14Scer\UAS.cLa.T:Disc\RFP at the A-P boundary by Scer\GAL4ptc.PU decreases distance between the L3 and L4 wing veins. Overexpression of mir-14Scer\UAS.cLa.T:Disc\RFP in the anterior wing compartment driven by Scer\GAL4ci.PU significantly reduces the size of the A compartment, causing it to curve in the anterior direction.

Animals expressing mir-14Scer\UAS.cLa.T:Disc\RFP under the control of Scer\GAL4da.G32 die at a late pupal stage with no obvious phenotypic defects in the pharate animal.

Expression of mir-14Scer\UAS.cLa.T:Disc\RFP under the control of either Scer\GAL4ptc-559.1 or Scer\GAL4Bx-MS1096 does not result in a mutant phenotype.

Expression of mir-14Scer\UAS.cLa.T:Disc\RFP under the control of Scer\GAL4sd-SG29.1 results in slightly curved wings.

Genetic Interactions
Statement
Reference

Co-expression of mir-14Scer\UAS.cLa.T:Disc\RFP suppresses the increased distance between the L3 and L4 wing veins seen in Scer\GAL4ptc.PU>ptcJF03223 flies; these flies even have a partial reduction in the distance between L3 and L4 (towards Scer\GAL4ptc.PU>mir-14Scer\UAS.cLa.T:Disc\RFP phenotypes, with a slight curving of the L4 vein towards the anterior portion of the wing).

Comments

Expression of mir-14Scer\UAS.cLa.T:Disc\RFP driven by Scer\GAL4btl.PU rescues the excess tracheal terminal cell phenotype seen in mir-14Δ1/mir-14Δ1 third instar larvae.
