Description

UAS regulatory sequences drive expression of a DsRed(Unk) fluorescent protein. A genomic fragment containing the mir-285 pre-miRNA hairpin has been inserted into the DsRed(Unk) 3'UTR.

The expression mir-285UAS.DsRed(Unk) under the control of Scer\GAL4ey.PU induces a small eye phenotype. Expression under the control of Scer\GAL4GMR.PU induces a rough eye phenotype associated with substantial apoptosis in the eye disc. Expression under the control of either Scer\GAL4hh.PU or Scer\GAL4en.PU induces wrinkled wings, associated with substantial apoptosis in the wing disc.

Expression under the control of Scer\GAL4moody.PU induces a compromised blood-brain barrier, as dye injected into the body cavity is able to diffuse into the adult retina or third instar larval brain. In agreement, larval subperineurial glial cells show disrupted/discontinuous septate junctions (Dlg immunostaining); these cells also show significant decreases in nuclear size and in ploidy only at higher expression conditions (i.e. at 25[o]C but not at 18[o]C), as compared to controls. There are also a significant increase in larval brain volume only at higher expression conditions (i.e. at 25[o]C but not at 18[o]C), as compared to controls.

Expression under the control of Scer\GAL4repo.PU results in a compromised larval blood-brain barrier, as injected dye into the body cavity is able to diffuse into the larval brain; no significant apoptosis is observed in the larval brain.

Adults expressing mir-285Scer\UAS.T:Disc\RFP under the control of Scer\GAL4Bx-MS1096 have curved wings.

Expression of mir-285Scer\UAS.T:Disc\RFP under the control of Scer\GAL4sd-SG29.1 results in curved and folded wings which are often broken.

Expression of mir-285Scer\UAS.T:Disc\RFP under the control of Scer\GAL4ptc-559.1 does not result in a mutant phenotype.

Expression of mir-285Scer\UAS.T:Disc\RFP under the control of Scer\GAL4Bx-MS1096 results in wing blisters.
