Description

257bp deletion between two divergently transcribed genes, CG14795 and inc. The transcription initiation site and 50bp of 5' untranslated region sequence of inc is removed by the deletion, while the 5' terminus and a small amount of upstream sequences of CG14795 remains intact. The level of both CG14795 and inc transcripts is reduced in the mutant.

Comment:

257 bp deletion removes a transcription start of inc and upstream region to CG14795.

Detailed Description

inc1 mutant adults sleep less (display reduced total amount of both daytime and nighttime sleep, along with short sleep bout length with increased bout number) than wild-type controls. Mutant third instar larvae show increased number of boutons on neuromuscular junction. Postsynaptic muscle recordings reveal altered synaptic transmission in inc1/incf00285 larvae: the amplitude of spontaneous miniature postsynaptic potentials is comparable to controls but their frequency is reduced, the amplitude of evoked potentials is decreased as is the quantal content.

These homozygotes are also short lived compared to controls.

The sleep deficit (reduced total amount of both daytime and nighttime sleep, along with short sleep bout length with increased bout number) characteristic for inc1 mutant adults can be partially rescued by Scer\GAL4inc.PS-driven expression of either Mmus\Kctd2Scer\UAS.T:Hsap\MYC or Mmus\Kctd5Scer\UAS.T:Hsap\MYC (but not Mmus\Kctd17Scer\UAS.3.T:Hsap\MYC or Mmus\Kctd17Scer\UAS.2.T:Hsap\MYC) in the mutant background.

Comments

The sleep deficit (reduced total amount of both daytime and nighttime sleep, along with short sleep bout length with increased bout number) characteristic for inc1 mutant adults is rescued by expression of either incScer\UAS.T:Hsap\MYC or incScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4inc.PS.
