The expression of Rab2HMS01271 under the control of Scer\GAL4SPARC-MI00329-GAL4 (and Dicer-2, for a more efficient RNAi) in the larval fat body induces defective secretion, including extracellular matrix material, as shown by the retention of both secr.GFP and Vkg.GFP. Expression under the control of Scer\GAL4Cg.PA alone leads to an increase in the size of the cis-Golgi (Grasp65.RFP) without a similar effect on endoplasmic reticulum exit sites (Sec16.GFP).
Expression of Rab2HMS01271 under the control of either Scer\GAL4Act5C.PU or Scer\GAL4Mef2.PU results in smaller synapse size and reduced number of boutons on neuromuscular junctions in third instar larvae compared to controls.

The neuromuscular junction (NMJ) structural defects (smaller synapse, reduced number of boutons) characteristic for third instar larvae expressing either Rab2HMS01271 or ICA69GD7020 under the control of Scer\GAL4Act5C.PU are not further worsened when the two RNAi alleles are co-expressed. However, the Rab2HMS01271 RNAi-induced NMJ defects can be rescued by co-expression of ICA69Scer\UAS.FL.
